Ok, yeah, puzzled here too.? You might try picking the receive audio off the DIN connector (packet, pins 5 & 2) just to see if it's present there.? That might help isolate what's happening.? I assume if you turn up the audio you can hear it in the radio's speaker.

Separately, I'm puzzled how you're getting good transmit audio when keying the rig with CAT.? You must also be keying with RTS or similar via the Data port, or are blasting the audio through (speaker-level).? In my measurements, there's something like 20db loss when feeding the audio in from the back, and keying the rig from CAT.

As I said in my mail, I have swapped the FT5000 onto the port I use on the interface for VHF and all works fine, so my assumption is the PC > Interface >Ft5000 is fine on either port of the interface, so the problem is in the lead (which I have replaced with no luck) or the data jack on the FT847,

There has been no hardware changes since day 1 set up of FT8 on 2m and all worked until recently.

I cant help thinking something has gone wrong on the data jack port, it all seems to point to that, anyone had similar problem?

Audio routing on the 847 is fixed, so a grayed-out config button is correct.? Receive audio goes to all the ports (Data, DIN, and the speaker), but transmit audio source is dependent on the mode and where PTT is triggered from.? SSB transmit audio comes from the Data jack; FM transmit audio comes from the DIN connector.? If you key the rig from CAT, the transmit audio comes from the mike connector on the front, which is probably the last place it would come from.

If your transmit audio went away I would suspect that you weren't in SSB mode, but that shouldn't affect receive.? Sounds to me like a problem on the PC side of things (audio mixer settings).
